Cultivation of leaf-tissue of Polyscias fruticosa (L.) Harms for quantity of saponin accumulation

Abstract

Polyscias fruticosa (L.) Harms is the medicinal plant, popularly cultivated in south-east asia and using as functional-foods. Besides of plantlets micropropagation for area development, hairy-root culture was the techniques manipulated for saponin accumulation. The leaves of young Dinh lang 1-year trees were used as culture materials. Sterilisation with javel water diluted to 50% in 10 minutes gave good results. MS medium supplemented with 1 mg/l NAA was favored for hairy-root induction from leaves tissue cultivation. Treatments with 2.4-D showed that hairy-root was not performed and induced callus strongly. Hairy-root was proliferation strongly in the MS medium supplemented with 0.5 mg/l NAA. Hairy-root cultivated on medium supplemented with IBA was weakly and thin in comparison with NAA. Kinetin was not affected on hairy-root proliferation. Quantity of oleanolic acid and saponin in proliferated hairy-root by HPLC method to show oleanolic acid 40,1 µg/g and saponin 396,2 µg/g accumulation.
