Abstract

In vitro leaves of Polyscias fruticosa L. Harms were induced callus on MS medium supplemented with 2,4D 2mg/l, kinetin 1mg/l, sucrose 30g/l gave callus initiation percentage of 94.63%. Media cultivation of MS supplemented with 2,4D 1mg/l, kinetin 0.5mg/l, sucrose 30 g/l, coconut water 10 % was favored for proliferation of callus to reach the growth rate of 19.83. Callus cells were white, soft with the high growth rate. Supplement with yeast extract, chitosan, casein hydrolysate separately to proliferated medium to show that the yeast extract 150 mg/l gave the highest saponin accumulation of 3480.2 µg/g and reasonable growth rate of 17.58.
